RESUMEN

White matter injury (WMI) is one of the most serious complications associated with preterm births. Damage to oligodendrocytes, which are the key cells involved in WMI pathogenesis, can directly lead to myelin abnormalities. L-ascorbyl-2-phosphate (AS-2P) is a stable form of vitamin C. This study aimed to explore the protective effects of AS-2P against chronic hypoxia-induced WMI, and elucidate the underlying mechanisms. An in vivo chronic hypoxia model and in vitro oxygen-glucose deprivation (OGD) model were established to explore the effects of AS-2P on WMI using immunofluorescence, immunohistochemistry, western blotting, real-time quantitative polymerase chain reaction, Morris water maze test, novel object recognition test, beaming-walking test, electron microscopy, and flow cytometry. The results showed that AS-2P resulted in the increased expression of MBP, Olig2, PDGFRα and CC1, improved thickness and density of the myelin sheath, and reduced TNF-α expression and microglial cell infiltration to alleviate inflammation in the brain after chronic hypoxia. Moreover, AS-2P improved the memory, learning and motor abilities of the mice with WMI. These protective effects of AS-2P may involve the upregulation of protein arginine methyltransferase 5 (PRMT5) and downregulation of P53 and NF-κB. In conclusion, our study demonstrated that AS-2P attenuated chronic hypoxia-induced WMI in vivo and OGD-induced oligodendrocyte injury in vitro possibly by regulating the PRMT5/P53/NF-κB pathway, suggesting that AS-2P may be a potential therapeutic option for WMI.

The presence of oil in coal seams from coal-oil symbiosis areas poses a serious threat to the safe and efficient mining of coal. However, the information about the application of microbial technology in oil-bearing coal seams was insufficient. In this study, the biological methanogenic potential of coal and oil samples in an oil-bearing coal seam was analyzed by anaerobic incubation experiments. The results showed that the biological methanogenic efficiency of the coal sample increased from 0.74 to 1.06 from day 20 to day 90, and the biological methanogenic potential of the oil sample was about twice as high as that of the coal sample after 40 days of incubation. The Shannon diversity and observed operational taxonomic unit (OTU) number of oil were lower than those in coal. The major genera in coal were Sedimentibacter, Lysinibacillus, Brevibacillus, etc., and the major genera in oil mainly included Enterobacter, Sporolactobacillus, and Bacillus. The methanogenic archaea in coal mainly belonged to the order Methanobacteriales, Methanocellales, Methanococcales, etc., and the methanogenic archaea in oil mainly belonged to the genera Methanobacterium, Methanobrevibacter, Methanoculleus, and Methanosarcina. In addition, metagenome analysis showed that functional genes belonging to processes such as methane metabolism, microbial metabolism in different environments, and benzoate degradation were in a higher abundance in the oil culture system, while genes belonging to sulfur metabolism, biotin metabolism, and glutathione metabolism were in a higher abundance in the coal culture system. The metabolites specific to coal samples mainly belonged to phenylpropanoids, polyketides, lipids, and lipid-like molecules; meanwhile, the metabolites specific to oil were mainly organic acids and their derivatives. In summary, this study has a reference value for the elimination of oil from coal in oil-bearing coal seams and can be used to separate oil from oil-bearing coal seams and reduce the hazard brought by oil for coal seam mining.

As novel optical nanoprobes, surface-enhanced Raman scattering (SERS) tags have drawn growing interests in the application of biomedical imaging and phototherapies. Herein, we demonstrated a novel in situ synthesis strategy for GO wrapped gold nanocluster SERS tags by using a tris(2,2'-bipyridyl)ruthenium(II) chloride (Rubpy)/GO nanohybrid as a complex Raman reporter, inspired by the role of GO as an artificial receptor for various dyes. The introduction of GO in the synthesis procedure provided systematic solutions for controlling several key parameters of SERS tags, including reproducibility, sensitivity, and colloidal and signal stability. An additional interesting thermal-sensitive SERS property (SERS intensity decreased upon increasing the temperature) was also achieved due to the heat-induced release/redistribution of reporter molecules adsorbed on GO. Combining the synergic effect of these features, we further fabricated multifunctional, aldehyde group conjugated Au@Rubpy/GO SERS tags for optical labeling and photothermal ablation of bacteria. Sensitive Raman imaging of gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli) bacteria could be realized, and satisfactory photothermal killing efficacy for both bacteria was achieved. Our results also demonstrated the correlation among the SERS intensity decrease ratio, bacteria survival rate, and the terminal temperature of the tag-bacteria suspension, showing the possibility to use SERS assay to measure antibacterial response during the photothermal process using this tag.
